If a spot opens in your party due to a death, that spot must be filled by a Pokemon from the PC immediately. This means the only way to use a new Pokemon is if one from your current party dies. You can't swap your party Pokemon for one from the PC. If a Pokemon faints, it's considered dead, and must be released/boxed permanently.
